Antibody Validation for Flow Cytometry

It was great to work with Rachel Leeson from Addgene to develop this short blog describing how to validate antibodies for flow cytometry. Key points:

  • Confirm/refute that your antibody can detect the antigen of interest, ideally at endogenously expressed levels, using your specific staining protocol.
  • Test several antibodies if possible.
  • When a good candidate(s) are found, explore their validity in your precious samples of interest.
  • This might require more than one approach if genetic manipulation of your cells of interest is not possible.

Improving Antibody Reproducibility with OGA

The Only Good Antibodies (OGA) community is collaborating with NC3Rs to address challenges in research reproducibility involving antibodies. Poorly characterized and inadequately performing antibodies lead to significant waste, including the unnecessary use of animals in their production and in research studies.

Earlier this year, we co-hosted a meeting with NC3Rs, bringing together antibody manufacturers, end-users, the pharmaceutical industry, academic researchers, publishers, research funders, and experts in research culture and integrity. Participants shared insights on challenges such as quality control, lot-to-lot variability, and the need for better validation practices, and explored actions to improve reproducibility and research integrity.

How MRC supports OGA community to drive improvements in antibody choice and use

The OGA team has received a prestigious Investigator Award from the MRC Better Methods Better Research Panel. This grant will support the OGA team in their work to improve the selection and use of high-quality antibodies in biomedical research, enhancing the integrity, reproducibility, and impact of research outcomes.

The project aims to benefit researchers, funders, biotech companies, and ultimately patients by reducing waste and ensuring meaningful, trustworthy scientific findings.
